Vergleich von Aufbereitungsverfahren zur Reinstwasserherstellung für die Produktion fester Arzneimittelformen unter Berücksichtigung der Lagerungs- und Verteilungssysteme (engl.: Comparison of treatment processes for the production of ultrapure water for the production of solid pharmaceutical forms, taking into account the storage and distribution systems)

Pharmawasser ist aufgrund seiner vielen Anwendungsbereiche in der Pharmaindustrie ein essentieller Stoff. Einem Pharmawassersystem, bestehend aus einer Herstellungsanlage sowie einem Lagerungs- und Verteilungssystem, kommt daher eine wichtige Bedeutung zu, um sowohl eine dem Arzneibuch entsprechende Qualität, aber auch Quantität sicherzustellen. Da die Herstellungsanlage eines bestimmten Pharmaunternehmens nicht mehr dem Stand der Technik entspricht, ist diese zu ersetzen. Für die Entwicklung eines Anlagenkonzeptes zur Herstellung von Gereinigtem Wasser, auch Purified Water genannt, wurde eine Literaturrecherche zu möglichen Behandlungsverfahren durchgeführt und das Pharmawassersystem des Pharmaunternehmens mit Hilfe einer Bedarfsermittlung analysiert. Für den Zeitraum einer Woche erfolgte die Ermittlung der Entnahmemengen und Volumenströme je Entnahmestelle. Es zeigte sich, dass die Herstellungskapazität der Neuanlage vergleichbar zur aktuellen Herstellungskapazität zu wählen ist. Eine Überlastung des Verteilungssystems konnte nachgewiesen werden. Mit Hilfe einer Abkopplung von Entnahmestellen, welche in einer separaten Ringleitung zusammengefasst werden, kann diese Überlastung behoben werden. Eine in die Bedarfsermittlung integrierte Prognose lässt den Schluss zu, dass die Lagerkapazität erweitert werden kann. Die entsprechende Qualität für Purified Water kann durch ein Anlagenkonzept, bestehend aus Grobfiltration, Ultrafiltration und Ionenaustauscher sowie Umkehrosmose, Membranentgasung und Elektrodeionisation sichergestellt werden. Innerhalb eines Lastenhefts werden die Anforderungen detailliert aufgeführt.
Pharmaceutical water is a major commodity in the pharmaceutical industry because of its application in many processes. A pharmaceutical water system consists of a treatment plant, a storage and distribution system and ensures a high water quality as well as quantity. As the treatment plant of a specific pharmaceutical company is not in accordance with the state of the art it is necessary to substitute it. In order to design a concept for a system of treatment steps required for production of purified water, a literature research about typical production methods is accomplished as well as an analysis of the current purified water consumption in the production processes. The aim of the latter was to determine the demand of purified water and the volume flow for each point of use during a period of one week. It was found that the production capacity of the existing plant is sufficiently dimensioned. Consequently, the
production capacity of the new treatment plant should not largely exceed the current capacity. Furthermore, this study revealed that the distribution system is overloaded. To reduce the load, a decoupling of some points of use is necessary. Therefore, they will form a new distribution loop in future. Considering the future water consumption, an expansion of the storage capacity
is required. The high quality of purified water could be reached by a treatment system consisting of prefiltration, ultrafiltration, ion exchange, as well as reverse osmosis, membrane degassing, and electrodeionisation. All requirements for the new treatment plant are summarized in a user requirement specification.
